Glioma is a common intracranial tumor, and the most malignant is glioblastoma. Abnormal rapid proliferation, strong invasiveness, and high heterogeneity are typical features of glioblastoma. The average median survival of patients is only about a year. Recently, the Zhao Xudong team of the Kunming Institute of Zoology of the Chinese Academy of Sciences and Huang Jingfei teamed up to make new progress in the glioblastoma typing research. The relevant research results were published in "Oncogenes".
One of the important reasons why glioblastoma is difficult to treat is its high degree of heterogeneity. However, the existing classification method based on histopathological features cannot explain a series of problems caused by tumor heterogeneity. For example, patients with the same pathological grade have different drug reactions and prognosis after the same treatment, so they need to be improved. For the diagnosis and treatment of glioblastoma, new classification methods need to be found.
Researchers have found that up to 60% of human glioblastoma samples have a CDKN2A gene deletion, and 30% of the samples have TP53 gene mutations, which are often "mutually exclusive". These two groups of patients have distinct genetic expression profiles and patient survival curves show significant differences. Glioblastoma typing with CDKN2A and TP53 gene mutations can cover 80% of glioblastoma samples with significant genetic representation.
The researchers successfully induced a glioblastoma model with typical pathological and molecular expression characteristics of human glioblastoma in mice. The successful construction of this animal model was based on CDKN2A and TP53 genes as characteristic genes. The typing of cell tumors provides reliable model support.
